01 — Overview

Description

A redox cofactor and bioactive compound that stimulates mitochondrial biogenesis through PGC-1α activation. The disodium salt is the stable, water-soluble form used in commercial supplement formulation.

Reddish-brown to dark red crystalline powder. Water-soluble. Stable through standard supplement-manufacturing temperatures when protected from prolonged light exposure.

02 — Background

Introduction

Pyrroloquinoline Quinone was first identified in 1979 as a bacterial cofactor for methanol dehydrogenase by J. A. Duine and colleagues in the Netherlands. The molecule has since been shown to occur in trace amounts in human breast milk, fermented foods, and a range of plant tissues.

Commercial production is by fermentation using Hyphomicrobium denitrificans or related methylotrophic bacteria, followed by extraction, chromatographic purification, and conversion to the disodium salt for stability and solubility in finished-product formulation.

Approved as a new dietary ingredient by the U.S. FDA under NDI 786 in 2008 (BioPQQ filing) and recognised as a novel food ingredient in the EU following EFSA review. Approved in Japan and China under their respective supplement frameworks.

PQQ acts both as a direct antioxidant capable of approximately 20,000 catalytic redox cycles per molecule (compared to roughly four for ascorbate) and as a signalling molecule that upregulates mitochondrial biogenesis through the PGC-1α and CREB pathways. The combination of these two mechanisms underpins its position in the premium longevity-supplement category.

Typical finished-product dosing is 10 to 20 mg per day, which makes PQQ one of the most cost-intensive ingredients in modern supplement formulation on a per-serving basis. The ingredient is almost always positioned in premium price tiers and is rarely included in mass-market multivitamins.
