01 — Overview

Description

Papain is a cysteine protease extracted from the latex of unripe papaya (Carica papaya) fruit. It is the original commercial meat tenderiser and one of the oldest enzymes in industrial food use, with broad endopeptidase activity across a wide pH range.

Light yellow to off-white free-flowing powder. Activity is standardised in USP Units, NF Units, TU (Tyrosine Units) or MCU (Milk Clotting Units) depending on the application and the buyer's specification.

02 — Background

Introduction

Papain has been used as a meat tenderiser by indigenous populations of Central and South America for at least 500 years, where unripe papaya was wrapped around tough cuts of meat before cooking. The active enzyme was characterised in the late 19th century and entered commercial production in the early 20th century.

Industrial production starts with the controlled scoring of unripe papaya fruit on the tree, collection of the exuded latex, drying and milling. The crude latex powder is then refined through dissolution, filtration and selective precipitation to yield purity grades from crude industrial through pharmaceutical. Major production regions include India, Sri Lanka, Uganda, Tanzania and the Democratic Republic of Congo.

The enzyme is Generally Recognized as Safe by the U.S. FDA, listed in the JECFA enzyme compendium, approved as a processing aid in the EU and listed in USP, EP and JP pharmacopoeias.

Mechanistically, papain is a cysteine endopeptidase that hydrolyses internal peptide bonds with a broad specificity favouring large hydrophobic residues at the P2 position. The enzyme is active over an unusually wide pH range from pH 3 to pH 9 and from 30 to 75 degrees Celsius, which makes it tolerant of varied processing conditions and explains its broad application footprint.

Strategic positioning is unusual: papain is one of the few major industrial food enzymes that is plant-derived rather than fermentation-derived, which positions it favourably for clean-label and natural-ingredient claims, although tropical-agriculture supply variability and tree-yield decline have driven sustained interest in microbial alternatives.
