Description

An essential amino acid required for protein synthesis and the precursor for histamine, carnosine, and other biologically important compounds. Used in pharmaceutical applications, infant formula, and as a buffer in biotechnology formulations.

Introduction

L-Histidine is an essential amino acid for infants (conditionally essential for adults) and serves as the precursor for several biologically important compounds: histamine (the neurotransmitter and immune mediator), carnosine (the muscle dipeptide), and the imidazole ring of nucleotide cofactors.

Industrial production proceeds by submerged aerobic fermentation using engineered Escherichia coli or Corynebacterium glutamicum strains.

Recognized as a permitted food ingredient by the U.S. FDA, the European Food Safety Authority, and equivalent regulators worldwide. Listed in BP, USP, EP, and JP pharmacopoeias.

The molecule's pKa near physiological pH (approximately 6.0) makes it valuable as a buffer in biotech and pharmaceutical applications, particularly for monoclonal antibody formulation where histidine buffer outperforms alternative buffers in protein stability across freeze-thaw and lyophilization cycles. This biotech application has grown substantially since 2010.
